摘要:N4-乙酰脫氧胞嘧啶(ac4C)最近被發現是mRNA上的一種新的修飾。ac4C修飾被證明是RNA穩定性、RNA翻譯和熱應激反應的關鍵調節因子。但其在真核生物mRNA上的存在仍存在爭議。在植物中,ac4C的存在、分布模式和潛在功能尚不清楚。在這里,我們報道了ac4C在擬南芥 mRNA上的存在。通過比較兩種ac4C測序方法,我們發現acRIP-seq適合植物ac4C測序,而ac4C-seq不適合。我們展示了通過acRIP-seq獲得的擬南芥mRNA中ac4C的轉錄組全圖譜。ac4C分布分析表明,擬南芥mRNA中ac4C富集在翻譯起始位點和翻譯結束位點附近。

RNA N4-cytidine acetylation in Arabidopsis thaliana
Abstract:RNA acetylation on N4-cytidine (ac4C) has recently been discovered as a novel modification on mRNA. The ac4C modification was shown to be a key regulator of RNA stability, RNA translation, and the thermal stress response. However, its existence on eukaryotic mRNA is still controversial. In plants, the existence, distribution pattern and potential function of ac4C are largely unknown. Here we report the presence of ac4C on the mRNA of both Arabidopsis thaliana . By comparing two ac4C sequencing methods, we found that acRIP-seq but not ac4C-seq was suitable for plant ac4C sequencing. We present transcriptome-wide atlases of ac4C in Arabidopsis thaliana mRNAs obtained by acRIP-seq. Analysis of ac4C distribution showed that the ac4C concentration in Arabidopsis mRNA was near the start and end sites of translation.
